GLP-1, GIP and glucagon: how do semaglutide, tirzepatide and retatrutide differ?
Three molecules often discussed together, but with different mechanisms, evidence and regulatory status. This educational guide does not recommend medications or doses.
What are GLP-1-based medicines?
They act on hormonal signaling involved in glucose regulation, appetite and gastric emptying. Approved uses vary by medicine, formulation and country.
Is semaglutide only for diabetes?
No. Semaglutide activates the GLP-1 receptor. Some formulations are authorized for type 2 diabetes, while others are authorized for chronic weight management in eligible patients. Not all formulations have the same approved indications.
How is tirzepatide different?
Tirzepatide activates both GIP and GLP-1 receptors. It can improve glucose control, increase satiety and delay gastric emptying, particularly early in treatment. It is not accurately described as merely controlling insulin spikes.
What is retatrutide?
Retatrutide is an investigational medicine that activates GIP, GLP-1 and glucagon receptors. Glucagon-receptor activity may contribute to energy expenditure, but an increase in resting metabolism is not guaranteed for every individual. It must not be presented as an approved treatment or offered outside applicable regulatory and research frameworks.
What are the side effects and precautions?
Gastrointestinal effects such as nausea, vomiting, diarrhea, constipation and abdominal discomfort are common with these classes. Important risks and warnings may include dehydration, gallbladder problems, pancreatitis and product-specific contraindications. A clinician must review medical history, other medicines and the approved product information.
Are these suitable for anyone who wants to lose weight?
No. Eligibility depends on diagnosis, medical history, body mass index, associated conditions and local approvals. Treatment does not replace nutrition, exercise or follow-up. Weight loss may also involve lean-mass loss, making strength and body-composition assessment relevant.
Which works best?
Study outcomes cannot be compared directly without considering differences in participants, doses, duration and trial design. Treatment selection, where authorized, requires individualized medical assessment.
Does THE CLINIK prescribe these medicines?
This article is not an offer and does not confirm availability of any of these products. Available care options can be discussed during an individual assessment, subject to applicable authorizations and requirements.
